GSK2256098 is an investigational small molecule inhibitor of focal adhesion kinase (FAK), a non-receptor protein tyrosine kinase involved in regulating cell migration, adhesion, proliferation, and survival. By inhibiting FAK activity, GSK2256098 disrupts integrin-mediated signaling and downstream pathways such as ERK/MAPK and PI3K/Akt. This mechanism impairs tumor cell migration, proliferation, survival, and angiogenesis. The drug has been primarily developed for the treatment of cancers with particular focus on meningiomas harboring NF2 mutations. Clinical studies have shown that it is well tolerated and may improve progression-free survival in patients with recurrent or progressive NF2-mutated meningiomas[1][2][6][10].
