Overview

GSK2292767 is a potent and selective small molecule inhibitor of phosphoinositide 3‑kinase delta (PI3Kδ), with high selectivity over other PI3K isoforms[1][5][7]. It was developed by GSK as a back-up compound to nemiralisib (GSK2269557) for the treatment of respiratory diseases such as asthma and chronic obstructive pulmonary disease (COPD)[5][7]. The drug has demonstrated durable anti-inflammatory activity in preclinical models of Th2-driven acute lung inflammation[3]. In human studies, it has been evaluated as an inhaled dry powder formulation for safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy smokers[3][4].
