Overview

GSK2315698 + GSK2398852 is a combination investigational therapy for systemic amyloidosis consisting of miridesap (GSK2315698), a small molecule serum amyloid P component (SAP) depleter that removes SAP from the circulation, and dezamizumab (GSK2398852), a humanized IgG1 monoclonal antibody targeting SAP. SAP stabilizes amyloid fibrils that accumulate in organs in amyloidosis, protecting them from breakdown. The therapeutic aim is to first deplete circulating SAP using miridesap, then target residual SAP within amyloid deposits with dezamizumab, enabling immune-mediated clearance of amyloid from organs. The approach leverages complement activation and macrophage recruitment to remove the amyloid material. Both agents were developed by GSK for systemic amyloidosis. Clinical trials evaluated safety, pharmacokinetics, and effect on organ amyloid load, but development was discontinued after early phase studies[1][3][5][7].
