Overview

GSK2339345 is an investigational inhaled small molecule developed by GSK as a voltage-gated sodium channel (VGSC) blocker for the treatment of chronic cough, specifically refractory or chronic idiopathic cough. Its mechanism of action involves antagonism of sodium channels in airway sensory nerves, aiming to reduce cough reflex sensitivity. Clinical studies showed that while the drug reached airway sensory nerves and was well tolerated, it did not demonstrate anti-tussive efficacy compared to placebo. Development for this indication has been discontinued[2][4][5].
