Overview

GSK2392019A is a recombinant protein subunit vaccine developed by GSK for the prevention of *Staphylococcus aureus* infections, particularly surgical site infections in patients undergoing elective spinal surgery. The vaccine is a four-component formulation (4C-Staph) comprising the antigens alpha-hemolysin (HlaH35L), the EsxA-EsxB fusion protein, ferric hydroxamate-binding lipoprotein (FhuD2), and the conserved surface protein Sta011. It is formulated with GSK's proprietary AS01 adjuvant system (specifically AS01B or AS01E) to enhance the immune response. Despite progressing to Phase 2 clinical trials, GSK discontinued the development of the vaccine after it failed to demonstrate sufficient efficacy in preventing *S. aureus* infections in the target surgical population.
