Overview

GSK2618960 is a humanized monoclonal antibody that targets the interleukin-7 receptor alpha subunit (CD127). By binding to IL-7Rα, it blocks interleukin-7 (IL-7) signaling, which modulates T cell activity and is implicated in various autoimmune diseases. The drug was developed by GSK for potential use in conditions such as primary Sjögren’s syndrome and multiple sclerosis. In clinical studies, GSK2618960 demonstrated effective blockade of IL‐7 receptor signaling with full target engagement and maximal inhibition of downstream STAT5 phosphorylation. It was generally well tolerated but showed high immunogenicity with persistent antidrug antibodies detected in most subjects. Development for multiple sclerosis was discontinued, and no further development has been reported for other indications[1][2][6][7].
