GSK2862277 is a fully human single domain antibody (dAb) fragment developed by GSK as a potent and selective antagonist of tumor necrosis factor receptor 1 (TNFR1). It was designed to inhibit TNFR1 signaling, thereby reducing inflammation and tissue injury associated with acute lung injury (ALI), acute respiratory distress syndrome (ARDS), and other inflammatory respiratory diseases. The drug was administered via inhalation or intravenous routes in clinical studies. Although it showed pharmacodynamic effects such as reduced biomarkers of lung permeability and inflammation, its development was discontinued after phase I/II trials due to unmet protocol criteria[2][3][4][5][6][8].
