Overview

GSK287615 is an investigational therapeutic protein subunit vaccine developed by GSK for the treatment of chronic hepatitis B virus (HBV) infection. It consists of recombinant hepatitis B surface antigen (HBsAg) formulated with a potent Adjuvant System, typically AS01B, which is designed to stimulate robust cellular immune responses, specifically CD8+ cytotoxic T lymphocytes (CTLs), to overcome immune exhaustion in chronic carriers. The vaccine was primarily developed as the protein-boost component of the heterologous prime-boost regimen GSK3528869A, which also utilized viral vector-based vaccines (ChAd154-HBV and MVA-HBV). The goal of this therapeutic approach was to achieve a functional cure, characterized by sustained HBsAg loss. However, GSK terminated the development of the GSK3528869A regimen in 2023 following a Phase 2 interim analysis that suggested the treatment was unlikely to meet its primary efficacy endpoints.
