Overview

GSK2878175 is a potent, orally administered small molecule inhibitor developed as a pan-genotypic non-nucleoside inhibitor of the hepatitis C virus (HCV) NS5B RNA-dependent RNA polymerase. It acts by binding to the enzyme's primer grip site (site IV), inhibiting viral replication across multiple HCV genotypes. The drug was investigated in both monotherapy and combination regimens for chronic hepatitis C infection. Clinical studies demonstrated significant reductions in HCV RNA levels with good tolerability and no major safety concerns identified during trials. Despite promising early results, development of GSK2878175 for hepatitis C was discontinued after Phase II clinical trials[1][2][3][7].
