Overview

GSK2981710 is a medium-chain triglyceride (MCT) formulation developed by GSK for the potential treatment of age-related cognitive decline and Alzheimer's disease. It functions as a ketogenic precursor, intended to elevate plasma levels of ketone bodies such as β-hydroxybutyrate (BHB) to provide an alternative fuel source for neurons in the presence of glucose hypometabolism. In clinical trials, the compound was administered at doses up to 30g/day. While it successfully increased BHB levels, it failed to demonstrate significant improvements in cognitive function or memory-related neuronal activity in healthy older participants. The development appears to have stalled following these results, with gastrointestinal side effects like diarrhea being commonly reported.
