Overview

GSK2983559 is an orally active and potent small molecule inhibitor of receptor-interacting protein serine-threonine kinase 2 (RIPK2). It was developed by GSK as a selective RIPK2 kinase inhibitor intended to block proinflammatory cytokine responses. The drug demonstrated activity in preclinical models and human inflammatory bowel disease explant samples by inhibiting the NOD1/NOD2-RIPK2 signaling pathway that leads to NF-κB activation and subsequent production of proinflammatory cytokines such as TNF-α, IL-1β, and IL-6. GSK2983559 entered phase I clinical trials for inflammatory bowel diseases but development was discontinued due to non-clinical toxicology findings and reduced safety margins[1][3][5][8].
