Overview

GSK3008348 is a potent and selective small molecule antagonist of the integrin αvβ6, developed as the first inhaled inhibitor targeting this integrin for the treatment of idiopathic pulmonary fibrosis (IPF)[1][4][5]. By binding with high affinity to αvβ6 on damaged lung epithelium, it inhibits activation of transforming growth factor beta (TGF-β), thereby reducing collagen production and potentially slowing or halting fibrotic progression[1][4]. The drug is administered via nebulization directly to the lungs. Preclinical studies demonstrated rapid absorption from the lung into systemic circulation and a receptor dissociation half-life of approximately 9 hours[1]. Clinical trials have shown that single doses are generally well tolerated in healthy participants[4]. Development was led by GSK (formerly GlaxoSmithKline) as an antifibrotic agent for IPF; however, clinical development appears to have been terminated after phase I trials[2].
