Overview

GSK3174998 is a humanized, agonistic IgG1 monoclonal antibody that targets and activates the OX40 receptor (also known as CD134), which is expressed on activated T cells. By binding to OX40, GSK3174998 stimulates the proliferation and survival of memory and effector T lymphocytes, enhancing T-cell-mediated immune responses against tumor cells and leading to tumor cell lysis. The drug was developed as an immunotherapy for advanced or recurrent solid tumors, head and neck cancer, and multiple myeloma. Its mechanism includes both direct activation of effector T cells and potential depletion of regulatory T cells depending on the antibody isotype. Development was led by GlaxoSmithKline (now known as GSK), sometimes in partnership with Merck for clinical trials[1][3][5][6].
