BOS-475 (also known as GSK3183475) is a small molecule inhibitor targeting the bromodomain and extraterminal domain (BET) family of proteins, specifically acting as a selective inhibitor of the second bromodomain (BD2). It was originally developed by GlaxoSmithKline and later advanced by Boston Pharmaceuticals. The drug is designed for topical administration and has been investigated primarily for the treatment of psoriasis and other inflammatory skin diseases. Its mechanism involves inhibition of BET proteins such as BRD2, which modulates multiple clinically validated inflammatory pathways including IL-12/IL-23-p40, Th17 cytokines, TNF-alpha, MCP-1, IL-6, CXCL10 & CXCL11—key mediators in psoriasis pathophysiology. Clinical development reached Phase 1 trials in plaque psoriasis but no further development has been reported[1][2][4].
