GSK3335065 is a small molecule inhibitor of kynurenine monooxygenase (KMO) that was developed by GSK for the treatment of acute pancreatitis. The drug acts by blocking KMO, thereby reducing levels of 3-hydroxykynurenine (3HK) and other related metabolites in the tryptophan pathway, which are elevated in patients with acute pancreatitis. The compound was administered intravenously in early clinical studies to assess safety, tolerability, pharmacokinetics, and pharmacodynamics. Although partial inhibition of KMO activity was observed in humans consistent with preclinical findings, development was discontinued after a serious adverse event occurred during a phase 1 trial[1][4][6].
