GSK3342830 is a novel injectable catechol-cephem (β-lactam) antibiotic developed for the treatment of infections caused by Gram-negative bacteria, including multidrug-resistant isolates. It features enhanced stability to β-lactamases and incorporates a catechol group on its side chain that acts as a siderophore mimetic. This design allows the drug to utilize bacterial iron transport systems for improved penetration into Gram-negative bacteria. Once inside, it inhibits bacterial cell wall synthesis by binding to penicillin-binding proteins. The drug was developed by GSK (formerly GlaxoSmithKline) and entered phase 1 clinical trials in 2017 but was discontinued in 2018 due to poor tolerability observed in multiple-dose studies[3][5][6][7].
