Overview

GSK3389404 is a liver-targeted antisense oligonucleotide developed for the treatment of chronic hepatitis B (CHB) virus infection. It is the N-acetyl galactosamine (GalNAc)-conjugated version of bepirovirsen (GSK3228836). The drug is designed to inhibit synthesis of hepatitis B surface antigen (HBsAg) and all other hepatitis B virus proteins by targeting a shared region present in all HBV mRNAs and pregenomic RNA. The GalNAc conjugation enhances delivery to hepatocytes. In clinical studies, GSK3389404 demonstrated dose-dependent reductions in HBsAg levels with an acceptable safety profile, though no effective dosing regimen was identified in Phase 2 trials.
