Overview

GSK3536852A is an investigational bivalent HIV-1 protein subunit vaccine candidate developed by GSK. It consists of two recombinant envelope glycoproteins (gp120) derived from HIV-1 Clade C strains, specifically C.1086 and TV1.C, which are prevalent in Southern Africa. The vaccine is formulated with GSK's proprietary **AS01B Adjuvant System**, a liposome-based formulation containing two potent immunostimulants: 3-O-desacyl-4'-monophosphoryl lipid A (MPL), which acts as a Toll-like receptor 4 (TLR4) agonist, and QS-21, a saponin purified from the bark of the *Quillaja saponaria* tree. The mechanism of action involves the induction of high-titer binding antibodies and robust CD4+ T-cell responses directed against the HIV-1 envelope to prevent viral entry and infection. GSK3536852A has been evaluated in Phase 1 and Phase 2 clinical trials, such as HVTN 120, both as a standalone regimen and in prime-boost combinations with other viral vectors.
