Overview

GSK356278 is a potent, selective, orally bioavailable, and brain-penetrant small molecule inhibitor of phosphodiesterase 4 (PDE4), developed by GSK. It demonstrates anxiolytic and cognition-enhancing effects in preclinical species without inducing typical PDE4 inhibitor side effects such as nausea and vomiting. The drug elevates cAMP levels in the central nervous system by inhibiting PDE4 activity, which is implicated in psychiatric and neurologic diseases. Preclinical studies showed a superior therapeutic index compared to other PDE4 inhibitors like rolipram and roflumilast. GSK356278 has been investigated for the treatment of anxiety disorders, depressive disorder (including major depressive disorder), Huntington's disease, and related neuropsychiatric conditions. Clinical development reached phase I trials evaluating safety, tolerability, pharmacokinetics, and brain target engagement[1][2][3][4][6].
