GSK3739937 (also known as VH3739937 or GSK'937) is an investigational small molecule HIV-1 maturation inhibitor developed for the treatment of HIV infection. It acts by inhibiting the final protease cleavage event between the capsid and spacer peptide 1 in the HIV-1 gag protein, thereby interfering with viral maturation and resulting in non-infectious virus particles. This mechanism is distinct from other classes of antiretroviral drugs. GSK3739937 demonstrates low nanomolar potency against multiple HIV-1 gag polymorphisms and has a high barrier to resistance except for certain mutations such as A364V. The drug features a long oral half-life (approximately 3 days), supporting less frequent dosing than once daily regimens. Clinical development is led by ViiV Healthcare, with phase 2 studies ongoing for HIV infection.
