GSK3745417 is an intravenously administered small molecule agonist of the stimulator of interferon genes protein (STING; also known as transmembrane protein 173 or TMEM173). It is being developed by GSK for its immunoactivating and antineoplastic activities. The drug activates the STING pathway, leading to induction of type I interferons and proinflammatory cytokines, which can stimulate anti-tumor immune responses. In addition to immune activation, preclinical studies have shown that GSK3745417 can directly induce apoptosis in acute myeloid leukemia (AML) cells, suggesting dual mechanisms of action—direct tumor cell killing and immune activation. The drug is currently in phase 1 clinical trials for relapsed/refractory acute myeloid leukemia (AML), high-/very high-risk myelodysplastic syndromes (MDS), and solid tumors[1][2][4][5][6][8].
