Overview

GSK3845097 is an autologous, genetically engineered **T-cell receptor engineered T-cell therapy** developed by Adaptimmune and formerly partnered with GSK. Patient T cells are modified to express an affinity-enhanced T-cell receptor recognizing the cancer-testis antigens NY-ESO-1 and LAGE-1a in the context of HLA-A*02, together with a dominant-negative transforming growth factor-beta receptor II designed to reduce suppressive transforming growth factor-beta signaling in the tumor microenvironment. The program was evaluated after lymphodepleting chemotherapy in HLA-A*02:01, HLA-A*02:05, or HLA-A*02:06-positive patients with antigen-positive advanced synovial sarcoma or myxoid/round cell liposarcoma.
