GSK3943104 (also identified by the clinical trial code GSKVX000000030918) is an investigational HSV-targeted immunotherapy being developed by GSK for the treatment of genital herpes. It is a recombinant protein subunit vaccine formulated with the AS01 adjuvant system. The immunotherapy is designed to elicit a strong T-cell and antibody response against herpes simplex virus type 2 (HSV-2), with the goal of reducing viral shedding and the frequency of symptomatic recurrences in individuals with established infection. The AS01 adjuvant, which contains the toll-like receptor 4 (TLR4) agonist monophosphoryl lipid A (MPL) and the saponin QS-21, is critical for enhancing the magnitude and durability of the cellular immune response. As of 2024, it is being evaluated in Phase I/II clinical trials for both therapeutic efficacy in infected adults and safety/immunogenicity in healthy seronegative individuals.
