GSK4011499 is an investigational, next-generation HIV-1 maturation inhibitor developed by ViiV Healthcare for the treatment of HIV-1 infection. It functions by binding to the HIV-1 Gag polyprotein, specifically at the junction between the capsid (CA) and spacer peptide 1 (SP1). This binding sterically hinders the HIV protease from performing the final cleavage step necessary for viral maturation. Consequently, the virus particles released from infected cells are immature and lack the ability to infect new cells. GSK4011499 is designed to have improved potency and a better resistance profile compared to earlier maturation inhibitors, potentially offering a new therapeutic option for patients with multi-drug resistant HIV.
