Overview
GSK4381562 is a fully human immunoglobulin G1 (IgG1) monoclonal antibody that targets poliovirus receptor-related immunoglobulin domain-containing protein (PVRIG; also known as CD112R), an inhibitory receptor expressed on natural killer (NK) cells and T cells. By binding to PVRIG and blocking its interaction with CD112—overexpressed on tumor cells—GSK4381562 is designed to enhance NK cell and T cell activation within the tumor microenvironment. This immune checkpoint inhibitor has potential antineoplastic activity by promoting both direct cytotoxicity via antibody-dependent cellular cytotoxicity (ADCC) and stimulation of innate and adaptive immune responses against tumors. The drug was originally developed by Surface Oncology as SRF813 before being licensed globally to GSK for further development[5][7][4][1].
