GSK4427296 is an investigational autologous T-cell receptor (TCR) therapy designed to target solid tumors expressing the cancer/testis antigen NY-ESO-1. The therapy involves engineering a patient's own T cells to express an affinity-enhanced TCR specific for NY-ESO-1, enabling these modified cells to recognize and bind to tumor cells that overexpress this antigen. Upon administration as an intravenous infusion, these engineered T cells seek out and attack NY-ESO-1-positive tumor cells. The product incorporates Lyell Immunopharma's Epi-R epigenetic reprogramming technology to enhance the stemness and functional persistence of the infused T cell population. It was developed in collaboration between GlaxoSmithKline (GSK) and Lyell Immunopharma[1][2][7][8].
