GSK5533524 is an investigational monoclonal antibody developed by GSK that acts as an agonist of the CD226 receptor (also known as DNAX Accessory Molecule-1 or DNAM-1). CD226 is a co-stimulatory protein expressed on the surface of Natural Killer (NK) cells and T cells, playing a crucial role in immune cell activation and tumor cell recognition. In the tumor microenvironment, the CD226 pathway is often suppressed by the presence of inhibitory receptors like TIGIT and PVRIG, which compete for the same ligands (such as PVR/CD155). By agonistically targeting CD226, GSK5533524 aims to bypass this inhibition and directly stimulate immune effector cells to enhance their anti-tumor activity. The drug is currently in Phase 1 clinical development for the treatment of advanced solid tumors, where it is being studied as a monotherapy and in combination with other anti-cancer therapies.
