Overview

GSK618334 is an investigational small molecule drug developed by GSK (GlaxoSmithKline) as a selective dopamine D3 receptor antagonist. It was designed to bind strongly to dopamine type 3 receptors in the human brain and was being explored as a potential innovative treatment for substance dependence and possibly other compulsive behavioral disorders. The mechanism of action involves blocking D3 receptors, which are implicated in addiction pathways; this blockade is hypothesized to reduce craving with potentially fewer side effects than existing therapies for addiction. Clinical development included several Phase 1 studies evaluating safety, tolerability, pharmacokinetics, and brain receptor occupancy in healthy volunteers[2][4][6].
