GSK706769 is an investigational, small-molecule HIV-1 maturation inhibitor developed by ViiV Healthcare, a joint venture led by GSK. It targets the final stage of the HIV-1 life cycle by binding to the Gag polyprotein at the junction between the capsid protein (CA) and spacer peptide 1 (SP1). This binding prevents the HIV-1 protease from cleaving the CA-SP1 site, which is a critical step for the formation of a mature, infectious viral core. As a result, the virus particles produced are immature and non-infectious. GSK706769 is designed to provide a new therapeutic option for individuals living with HIV, including those who may have developed resistance to other classes of antiretroviral therapy. It is currently being evaluated in early-phase clinical trials to assess its safety, tolerability, and pharmacokinetics.
