GSK856 is a novel, small-molecule dimeric amidobenzimidazole that acts as a Stimulator of Interferon Genes (STING) agonist. Developed by GSK, it is designed for systemic (intravenous) administration to enhance antitumor immunity by inducing the expression of interferon-stimulated genes (ISGs) and the production of proinflammatory cytokines. In veterinary clinical studies involving dogs with various solid tumors, including malignant melanoma and soft tissue sarcoma, GSK856 demonstrated on-target pharmacodynamic effects and a manageable safety profile, characterized by transient cytokine release consistent with activation of the STING pathway.
