Overview

GST-HG131 is a novel, orally bioavailable small molecule inhibitor developed for the treatment of chronic hepatitis B virus (HBV) infection. It is a dihydroquinolizinone (DHQ) compound that acts by inhibiting the expression and secretion of hepatitis B surface antigen (HBsAg) and e-antigen (HBeAg), which are key viral proteins involved in HBV persistence. The drug exerts its antiviral effect primarily through binding to and inhibition of Terminal nucleotidyltransferase 4B (TENT4B), an enzyme responsible for polyadenylation of HBV mRNAs; this leads to shortening of the poly-A tail, accelerating degradation of viral mRNA and reducing antigen production[2][6]. Preclinical studies demonstrated potent activity in vitro and in vivo with improved safety over earlier compounds such as RG7834[2][8]. Clinical trials have shown that GST-HG131 is generally well tolerated at certain doses, with ongoing development focused on chronic hepatitis B[7].
