GST-MDA-7 is a recombinant fusion protein consisting of the cytokine Melanoma Differentiation Associated gene-7 (MDA-7), also known as Interleukin-24 (IL-24), fused to a Glutathione S-transferase (GST) tag. MDA-7/IL-24 is a unique member of the IL-10 cytokine family that demonstrates potent and selective anti-tumor activity by inducing apoptosis in a broad spectrum of cancer cells while remaining non-toxic to normal cells. The GST-tagged version is frequently utilized in laboratory research to elucidate the biochemical signaling pathways of the cytokine. Its mechanism of action involves the induction of endoplasmic reticulum (ER) stress and the activation of the CD95 (Fas) death receptor pathway through a ceramide-dependent mechanism. This leads to the activation of pro-apoptotic signaling molecules such as PERK, p38 MAPK, and JNK, while simultaneously inhibiting survival signals like ERK 1/2, ultimately resulting in caspase-mediated cell death.
