GSTP1 shRNA is a short hairpin RNA (shRNA) therapeutic candidate designed to silence the expression of Glutathione S-transferase P1 (GSTP1). GSTP1 is a Phase II metabolic enzyme often overexpressed in various malignancies, including glioblastoma multiforme (GBM), where it is associated with chemotherapy resistance and aggressive tumor growth. By utilizing a lentiviral vector to deliver the shRNA, the treatment achieves a significant knockdown of GSTP1 protein levels. This silencing leads to inhibited cell proliferation, induction of apoptosis via caspase 3/7 activation, and modulation of the MAPK signaling pathway, specifically increasing the phosphorylation of ERK1/2 and JNK1/2. Research conducted at Duke University suggests that GSTP1 is obligately required for the survival of high-GSTP1-expressing glioma cells, providing a rationale for its use as a targeted therapy.
