GT-00AxIL15 is a tumor-targeted interleukin-15-based immunocytokine that fuses a humanized IgG1 monoclonal antibody against a tumor-associated, glycosylated epitope of MUC1 (TA-MUC1) with IL-15 modules to deliver IL-15 signaling selectively to the tumor microenvironment.[1] The construct retains wild-type IL-15 receptor binding, engaging IL-15Rα/IL-15Rβγ on immune cells while simultaneously binding TA-MUC1 on tumor cells and Fcγ receptors via its Fc domain, thereby activating and expanding NK, NKT, CD8+ and CD4+ T cells, enhancing antibody-dependent cellular cytotoxicity, and driving anti-tumor cytotoxicity.[1] Developed by Glycotope, with clinical collaboration by LegoChem Biosciences, GT-00AxIL15 is being advanced for TA-MUC1-positive solid tumors with high unmet need, including ovarian, lung, and breast cancers, and is also being explored in combination with iPSC-derived NK cell therapy for triple-negative breast cancer.[1][3][4][7]
