Overview

GT-02037 is a glycosylated somatostatin analog (SSA) developed by GlyTech, Inc. for the treatment of acromegaly caused by pituitary adenomas. The drug is engineered using GlyTech's proprietary chemoenzymatic synthesis platform, which allows for the precise attachment of specific, well-defined glycans to the peptide backbone. This glycosylation is designed to optimize the drug's pharmacokinetic properties, potentially offering a longer duration of action or improved safety profile compared to conventional non-glycosylated somatostatin analogs like octreotide. GT-02037 acts as a selective agonist of somatostatin receptors, primarily the SSTR2 subtype, which are highly expressed on growth hormone-secreting pituitary adenoma cells. By activating these receptors, GT-02037 inhibits the hypersecretion of growth hormone (GH) and subsequently reduces levels of insulin-like growth factor 1 (IGF-1). It has been evaluated in early-phase clinical trials in Japan, including crossover studies comparing its efficacy and safety directly to subcutaneous octreotide.
