Overview

GT-03842 is a preclinical allosteric small molecule inhibitor of discoidin domain receptor 2 (DDR2), developed by Gain Therapeutics. Identified using the company’s Magellan Drug Discovery Platform, GT-03842 inhibits DDR2 phosphorylation and activation in a dose-dependent manner without targeting the kinase domain, distinguishing it from traditional kinase inhibitors. This novel mechanism may help circumvent resistance mechanisms commonly associated with kinase inhibitors and potentially offer improved selectivity and safety. Aberrant DDR2 activity is implicated in various cancers, including metastatic breast cancer, where it drives tumor progression, proliferation, migration, invasion, metastasis, epithelial-mesenchymal transition (EMT), and immunotherapy resistance. Preclinical data show that GT-03842 reduces DDR2 phosphorylation both in vitro (HEK293 cells) and in vivo (metastatic breast cancer models)[1][2][3][4].
