GT-04686 is an oral, brain-penetrant small molecule allosteric regulator of the enzyme glucocerebrosidase (GCase), developed by Gain Therapeutics. Discovered using the proprietary SEE-Tx® discovery platform, the compound is designed to act as a pharmacological chaperone that stabilizes the GCase enzyme, facilitating its proper folding and trafficking to the lysosome. By restoring GCase activity, GT-04686 aims to reduce the accumulation of toxic substrates such as glucosylsphingosine (GluSph) and glucosylceramide, which are implicated in lysosomal dysfunction and neurodegeneration. The compound is currently in preclinical development as a follow-on or next-generation candidate for the treatment of GBA1-associated Parkinson's disease and is being positioned for IND-enabling studies.
