Drug intelligence / Profile preview
GT-73 is a novel, small-molecule inhibitor of leukocyte transendothelial migration that acts by targeting platelet endothelial cell adhesion molecule-1 (PECAM-1/CD31). Developed by researchers at Bar-Ilan University and its spinout Silverskate Bio, GT-73 covalently binds to PECAM-1, preventing its homophilic dimerization and thereby blocking the migration of white blood cells across the endothelial barrier into tissues. This mechanism of action allows GT-73 to selectively inhibit the infiltration of inflammatory cells into end-organs without causing systemic immunosuppression. Preclinical studies have demonstrated the therapeutic potential of GT-73 in various animal models of autoimmune and inflammatory diseases, including neuropsychiatric systemic lupus erythematosus (NPSLE), acute respiratory distress syndrome (ARDS), multiple sclerosis, Crohn's disease, rheumatoid arthritis, and non-alcoholic fatty liver disease.
