GT20029 is a first-in-class, topical small molecule androgen receptor (AR) degrader developed using the proteolysis-targeting chimera (PROTAC) platform. Unlike traditional AR antagonists that block the receptor, GT20029 binds to and recruits AR proteins to E3 ubiquitin ligase, leading to their targeted degradation via ubiquitination. This mechanism reduces androgen sensitivity in local skin tissues such as hair follicle sebaceous glands, thereby inhibiting follicular miniaturization and promoting hair growth. The drug is designed for local action with minimal systemic absorption, reducing the risk of side effects associated with oral anti-androgen therapies. Developed by Kintor Pharmaceutical, GT20029 is primarily being investigated for the treatment of androgenetic alopecia (pattern hair loss), with additional consideration for acne vulgaris[1][2][3][4][5][6][7][8].
