GTB-3550 TriKE is a first-generation tri-specific killer engager (TriKE) developed for the treatment of relapsed/refractory acute myeloid leukemia (AML), high-risk myelodysplastic syndromes (MDS), and other CD33-positive hematologic malignancies. It is a single-chain recombinant fusion protein composed of the variable regions of anti-CD16 and anti-CD33 antibodies linked to a modified form of human interleukin 15 (IL-15). The molecule simultaneously binds CD16 on natural killer (NK) cells and CD33 on tumor cells while delivering an IL-15 signal to activate and expand NK cells. This design aims to enhance NK cell-mediated cytotoxicity against cancer cells without requiring exogenous cell therapy or combination drugs. Clinical studies demonstrated reductions in bone marrow blast levels in AML/MDS patients with good tolerability; however, development was discontinued in favor of more advanced second-generation molecules[1][3][5][6][7][9].
