Overview

GTB-4550 is a trispecific killer cell engager (TriKE) developed by GT Biopharma for the treatment of solid tumors, particularly those expressing PD-L1. The molecule is designed to simultaneously target three components: CD16a on natural killer (NK) cells, interleukin 15 receptor (IL15R), and programmed death-ligand 1 (PDL1) on tumor cells. By engaging CD16a, it activates NK cells; through IL15R agonism, it stimulates NK cell proliferation and persistence; and by inhibiting PDL1, it blocks an immune checkpoint pathway that tumors use to evade immune detection. This multi-targeted approach aims to enhance anti-tumor immunity by both activating and sustaining NK cell responses while overcoming tumor-mediated immunosuppression[1][7][9].
