Overview

GTB-7550 is a tri-specific killer engager (TriKE) developed by GT Biopharma for the treatment of CD19-positive lymphoid malignancies and autoimmune diseases. It is a recombinant fusion protein composed of three functional domains: - A camelid nanobody that binds to the CD16 receptor on natural killer (NK) cells, - The single-chain variable fragment (scFv) of an anti-CD19 antibody targeting B cells, - Human wild-type interleukin 15 (IL-15), which stimulates NK cell proliferation and activation. The mechanism leverages antibody-dependent cellular cytotoxicity by engaging NK cells via CD16, directing them to kill CD19-expressing malignant or autoreactive B cells. Preclinical studies have demonstrated activity against lymphoma and chronic lymphocytic leukemia models as well as potential in autoimmune disease settings such as lupus. As of late 2024, it remains in preclinical development with plans for clinical material production and regulatory submission after 2026[1][2][3][7][8].
