GTI-850 is a novel small molecule inhibitor of S-nitrosoglutathione reductase (GSNOR), developed by GSNO Therapeutics. It is designed to modulate nitrosylation signaling by inhibiting GSNOR, thereby increasing levels of endogenous S-nitrosoglutathione (GSNO). This mechanism leads to the suppression of autoimmunity, pro-inflammatory mediators, reactive oxygen/nitrogen species (ROS/RNS), matrix metalloproteinases (MMPs), tumor necrosis factor-alpha (TNF-α) and other cytokines, as well as pro-inflammatory B and T cells. Preclinical studies have demonstrated strong activity in models of rheumatoid arthritis and inflammatory bowel disease, with additional potential in age-related diseases such as periodontal disease and hearing loss. GTI-850 has shown a favorable safety profile in sub-chronic toxicity studies and exhibits oral bioactivity despite low oral bioavailability due to its local action in the intestine leading to systemic effects via increased GSNO[1][4][5].
