Overview

Guanoclor is a sympatholytic, peripherally acting antiadrenergic agent classified as an antihypertensive drug. It was developed by Pfizer and has been investigated for the treatment of various types of hypertension, including essential (unknown etiology), renal, and malignant hypertension. Guanoclor acts primarily as an adrenergic neuron-blocking agent and also inhibits noradrenaline synthesis by blocking the enzyme dopamine beta-hydroxylase. This results in reduced levels of noradrenaline and decreased blood pressure. Additionally, it binds to non-adrenergic sites in pig kidney membranes and has affinity for the Na+/H+ exchanger at a site distinct from that recognized by amiloride[1][3][4]. Guanoclor is a small molecule belonging to the dichlorobenzene class of organic compounds[2]. Its maximum clinical trial phase reached was Phase II[3].
