Overview

Guanoxan is a small molecule antihypertensive agent that was used for the treatment of hypertension. Its mechanism of action involves blocking alpha-2 adrenergic receptors (adrenoceptors), which inhibits the release of norepinephrine—a neurotransmitter that promotes vasoconstriction and increases blood pressure. By reducing norepinephrine release, guanoxan lowers both systolic and diastolic blood pressure in both lying and standing positions. Guanoxan is structurally classified as a benzo-1,4-dioxane derivative. The drug was approved in the UK but was withdrawn from the market due to hepatotoxicity (liver damage)[1][2][3][5][7].
