Overview

GUCY2C TAC (also known as GUCY2C-TAC T) is an investigational autologous T-cell therapy developed by Triumvira Immunologics for the treatment of colorectal cancer. The therapy utilizes the proprietary T-cell Antigen Coupler (TAC) technology platform, which genetically modifies a patient's own T-cells to express a TAC molecule designed to specifically target guanylyl cyclase 2C (GUCY2C), a receptor frequently overexpressed in colorectal cancer cells. Unlike traditional Chimeric Antigen Receptor (CAR) T-cells, TAC-T cells are engineered to activate the natural T-cell receptor (TCR) complex by co-opting the endogenous CD3 epsilon subunit. This mechanism is intended to provide a more physiological and controlled T-cell activation, potentially leading to sustained anti-tumor activity with a reduced risk of off-target toxicities. Preclinical data presented at SITC meetings in 2022 and 2023 demonstrated that GUCY2C TAC effectively eliminates GUCY2C-expressing tumor cells in vitro and shows robust anti-tumor efficacy in vivo.
