Gumetinib + toripalimab is a combination therapy comprising gumetinib, a highly selective and potent small-molecule MET inhibitor, and toripalimab, a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. Developed through a collaboration between Haihe Biopharma and Junshi Biosciences, this regimen is primarily investigated for the treatment of advanced non-small cell lung cancer (NSCLC) harboring MET alterations, such as MET exon 14 skipping mutations or MET amplification. The rationale for the combination lies in the potential synergy between MET inhibition and immune checkpoint blockade; MET signaling is known to contribute to an immunosuppressive tumor microenvironment and can drive resistance to PD-1/PD-L1 inhibitors. By simultaneously blocking MET-driven oncogenic signaling and restoring T-cell anti-tumor activity, the combination aims to improve clinical outcomes in patients with MET-dysregulated lung cancer.
