Overview

**GUT-108** is a rationally designed live biotherapeutic product consisting of an 11-strain bacterial consortium derived from healthy human stool isolates, optimized as a refined version of the earlier GUT-103 (17 strains) to treat inflammatory bowel disease (IBD). Developed by Gusto Global in collaboration with researchers at the University of North Carolina and the Crohn's and Colitis Foundation, it addresses dysbiosis in IBD patients by engrafting in the inflamed gut, reducing pathobionts (e.g., Enterobacteriaceae), expanding protective bacteria, and producing key metabolites such as butyrate, propionate, GABA, indole, and secondary bile acids (LCA, DCA) via enzymes like 7-α-dehydratase (7-α-DH), choloyl-CoA hydrolase (CGH), and others. In humanized mouse models of chronic T-cell-mediated colitis, therapeutic oral administration reversed established inflammation, decreased Th1/Th17 cells and cytokines (IFNγ, IL-17α, TNFα), increased IL-10-producing regulatory T cells, B cells, and dendritic cells, restored mucosal healing, and upregulated homeostatic pathways (e.g., IL-15, GPR41/43, FXR).[1][2][4][5]
