Overview
Gutrif is a generic formulation of rifaximin, a non-systemic, broad-spectrum rifamycin-derived antibiotic. Marketed by Rhombus Pharma, Gutrif is designed to act locally within the gastrointestinal tract due to its extremely low systemic absorption (<0.4%). Rifaximin exerts its bactericidal activity by binding to the beta-subunit of bacterial DNA-dependent RNA polymerase (rpoB), thereby inhibiting bacterial RNA synthesis and transcription. Additionally, it acts as an agonist of the human pregnane X receptor (PXR), which may contribute to anti-inflammatory effects in the gut mucosa. Gutrif is primarily indicated for the treatment of irritable bowel syndrome with diarrhea (IBS-D), travelers' diarrhea caused by non-invasive strains of Escherichia coli, and for reducing the risk of overt hepatic encephalopathy recurrence in adults.
