GV1001 + gemcitabine is a combination therapy consisting of **GV1001**, a telomerase-based cancer vaccine comprised of a 16-mer peptide derived from human telomerase reverse transcriptase (TERT), and **gemcitabine**, a nucleoside analog chemotherapeutic agent. GV1001 acts primarily by eliciting immune responses against tumor cells expressing telomerase, while gemcitabine impairs DNA synthesis in rapidly dividing cancer cells. When combined, GV1001 has shown synergistic effects with gemcitabine in preclinical and clinical studies of pancreatic ductal adenocarcinoma (PDAC), enhancing tumor cell death and reducing stromal fibrosis, thereby improving drug delivery. The combination may suppress pro-inflammatory cytokines involved in tumor microenvironment fibrosis, such as TNF-α, IL-6, and IL-1β, and may potentiate anti-cancer immunity. GV1001 additionally exhibits anti-angiogenic, anti-inflammatory, and immunomodulatory activities. Clinical studies indicate improvement in overall survival (OS) and time to progression (TTP) versus gemcitabine-based regimens alone in certain PDAC patient subgroups[1][2][3][4].
